Internal Standard Quantification

Internal standard quantification is an analytical method that determines the amount of a target compound by comparing its measured signal with that of a known reference, called an internal standard. The reference is added at a consistent amount to each sample and undergoes similar preparation, separation, and detection as the analyte, allowing the analyte-to-standard signal ratio to compensate for variations in sample handling and instrument response. In biology, this approach supports reliable measurement of metabolites, proteins, nucleic acids, and other biomolecules. It improves precision across samples and helps researchers compare biological conditions, assess experimental changes, and validate quantitative assays.

We describe a method to sort single mammalian cells and to quantify the expression of up to 96 target genes of interest in each cell. This method includes the use of internal qPCR standards to enable the estimation of absolute transcript counts.
