Iron Regulatory Proteins

Iron regulatory proteins (IRP1 and IRP2) are RNA-binding factors that maintain cellular iron homeostasis by coordinating the production, storage, and uptake of iron. When iron is scarce, they bind iron-responsive elements (IREs) in target messenger RNAs: binding near the 5′ untranslated region represses ferritin translation, while binding in the 3′ untranslated region stabilizes transferrin receptor mRNA and promotes iron uptake. When iron levels rise, IRP1 acquires an iron-sulfur cluster and functions as aconitase, whereas IRP2 is targeted for degradation, releasing these regulatory effects. Studying IRPs clarifies mechanisms underlying anemia, neurodegeneration, infection, and iron-related disease.

This article demonstrates how to prepare and administer transferrin-bound nonradioactive isotopic iron for studies of iron transport in mouse pregnancy. The approach for quantifying isotopic iron in fetoplacental compartments is also described.
