Gfp Fusion Screening

GFP fusion screening is a molecular biology technique that uses green fluorescent protein (GFP) linked to a target protein to identify and evaluate recombinant cells or constructs. After genetic fusion and expression, GFP fluorescence provides a visible readout of protein production, while microscopy or fluorescence measurements can reveal cellular localization, expression levels, and, in some cases, solubility. Researchers use this approach to screen transformed cells, select successful cloning or expression conditions, and track protein behavior in living cells. Because the signal can be monitored without destructive staining, GFP fusions support studies of gene expression, protein trafficking, cellular organization, and functional protein design.

Mitochondrial fusion was measured by tracking the equilibration of photoconverted matrix-targeted GFP across the mitochondrial network over time. Thus far, only one cell could be subjected to an hour long kinetic analysis at a time. We present a method that simultaneously measures multiple cells, thereby speeding up the data collection process.
