Kidney Slice Imaging

Kidney slice imaging is a microscopy-based technique for examining the structure and activity of kidney tissue within thin, prepared sections. By cutting viable or fixed kidney slices and imaging them with bright-field, fluorescence, or other optical methods, researchers can resolve nephron organization, cellular morphology, and spatial relationships while preserving tissue architecture. The approach supports studies of filtration, tubular transport, vascular organization, and responses to injury or pharmacological treatment. Because kidney slices provide a tissue-level context between isolated cells and whole-animal experiments, they help investigate renal physiology, disease mechanisms, and therapeutic effects with greater spatial and biological relevance.

Red marks all bacteria, and green highlights infection-responsive cells.

A Whole-Mount Organ Staining Technique for Three-Dimensional Imaging of the Kidney

2025

This video demonstrates an assay for the whole-mount staining of murine kidneys using a tissue-clearing method called CUBIC (clear, unobstructed brain/body imaging cocktails and computational analysis) and immunofluorescent staining. The kidney is subjected to tissue clearing, wherein the kidney is delipidated, decolorized, and treated with a refractive index matching solution to make the tissue transparent for optimum three-dimensional imaging. The tissue is stained with fluorescently labeled...

In this video, we demonstrate the vibratome sectioning of a whole mouse kidney to obtain thin slices. These sliced tissue sections can be stored at low temperatures until further downstream analysis.

Preparation of Acute Subventricular Zone Slices for Calcium Imaging

A method to load subventricular zone (SVZ) cells with calcium indicator dyes for recording calcium activity is described. The postnatal SVZ contains tightly packed cells including neural progenitor cells and neuroblasts. Rather than using bath loading we injected the dye by pressure inside the tissue allowing better dye diffusion.
