Myogenic Progression

Myogenic progression is the stepwise process by which progenitor cells commit to a muscle lineage, differentiate, and mature into functional muscle fibers. It begins with activation of myogenic regulatory factors such as MyoD and Myf5, followed by myogenin-driven differentiation, alignment, and fusion of myoblasts into multinucleated myotubes that organize contractile proteins into sarcomeres. In biology, studying this progression clarifies how skeletal muscle develops, repairs injury, and responds to disease or genetic disruption. These insights support research on muscle disorders, regenerative medicine, drug testing, and engineered muscle tissues.

Fabrication of Myogenic Engineered Tissue Constructs

Here, we demonstrate fabrication of collagen-based, tissue constructs containing skeletal myoblasts. These 3-D engineered constructs may be used to replace or repair tissues in vivo. For our purposes, we have designed these as an atrioventricular electrical conduit for the repair of complete heart block[1].

Pressure myography is used to assess vasoactivity of small arteries that develop sustained constriction when pressurized. This manuscript provides a detailed protocol to assess in isolated segments of small mesenteric arteries from rats, vasoactivity and the effect of intraluminal pressure on vascular diameter.
