Lactate Dehydrogenase

Lactate dehydrogenase (LDH) is an enzyme that catalyzes the reversible conversion of pyruvate to lactate, a reaction central to cellular energy metabolism. During anaerobic glycolysis, LDH uses NADH to reduce pyruvate to lactate while regenerating NAD+, allowing glycolysis to continue when oxygen availability or mitochondrial oxidation is limited. LDH exists as tissue-associated isoenzymes with distinct distributions, and their activity helps researchers study metabolic states and cellular injury. Measuring LDH in blood or cultured cells can support investigations of tissue damage, cancer metabolism, exercise physiology, and the balance between aerobic and anaerobic energy production.

A Lactate Dehydrogenase Release Assay To Detect Macrophage Death Following Nigericin Exposure

2025

This video demonstrates the lactate dehydrogenase assay to detect inflammasome-mediated macrophage death. The macrophages are exposed to lipopolysaccharide for priming, followed by exposure to nigericin to cause inflammasome-induced cell death, which is determined by the lactate dehydrogenase assay.

A Lactate Dehydrogenase Release Assay to Detect Cryptococcus neoformans Infection in Macrophages

This video describes a colorimetric assay for lactate measurement in cell lysates by using the lactate dehydrogenase enzyme. This enzyme converts lactate into pyruvate, along with the conversion of NAD to NADH that reacts with tetrazolium dyes and gives a colored product that can be measured spectrophotometrically to quantitate the lactate.
