Liver Engraftment

Liver engraftment is the establishment, survival, and functional integration of transplanted liver cells or tissue within a recipient’s liver, a process central to regenerative medicine and transplantation biology. After delivery, donor cells must adhere to the hepatic microenvironment, access blood flow, evade or withstand immune-mediated injury, and proliferate or repopulate damaged tissue while contributing to liver function. Engraftment is studied in hepatocyte transplantation, stem cell and organoid therapies, and experimental models of liver repair. Measuring cell retention, vascular integration, and restoration of metabolic activity helps researchers evaluate treatment effectiveness and improve strategies for treating liver failure and inherited metabolic disorders.

In Vivo Imaging of Liver Spheroids Engrafted in the Anterior Chamber of the Mouse Eye

Here, we describe a platform that allows noninvasive in vivo imaging of liver spheroids engrafted in the anterior chamber of the mouse eye. The workflow spans from generating spheroids from primary liver cells to transplantation into the mouse eye and in vivo imaging at cellular resolution by confocal microscopy.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.

Tracking Dynamics of Muscle Engraftment in Small Animals by In Vivo Fluorescent Imaging

We describe an in vivo fluorescence imaging protocol to monitor muscle regeneration by GFP-labeled myoblasts after transplantation into skeletal muscles of both healthy and dystrophic mice. This protocol can be adapted to study muscle regeneration by transplantation of other types of cells and in other muscular conditions as well.
