Luciferin Injection

Luciferin injection is a method for introducing the light-emitting substrate luciferin into a living organism or experimental model, enabling measurement of luciferase activity in biology. After administration, luciferin reaches cells expressing a luciferase enzyme; in the presence of oxygen and, for firefly luciferase, ATP, the enzyme catalyzes luciferin oxidation and produces visible or detectable photons. Researchers capture this bioluminescent signal with specialized cameras to track gene expression, monitor cell survival or movement, and evaluate biological responses over time. Because the signal can be measured noninvasively, luciferin injection supports longitudinal studies in molecular biology, disease models, and therapeutic research.

This manuscript provides the detailed procedure of intra-iliac artery (IIA) injection, a technique to deliver cancer cells specifically to hind limb tissues including bones to establish experimental bone metastases. Although initially established with breast tumor models, this protocol can be easily extended to other cancer types.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.
