Propidium Iodide Nuclear Staining

Propidium iodide nuclear staining is a fluorescence-based method for visualizing and quantifying cellular DNA, making it useful for assessing nuclear content and cell populations in biology. Because propidium iodide cannot readily cross intact plasma membranes, fixation or membrane permeabilization allows the dye to enter cells, where it intercalates into nucleic acids and emits red fluorescence when excited. RNase treatment can reduce RNA-associated signal and improve DNA-content measurements. Researchers use this technique in microscopy and flow cytometry to identify nuclei, evaluate membrane damage and cell viability, and analyze cell-cycle distribution, including the proportions of cells in G0/G1, S, and G2/M phases.
