Mammalian Expression Vectors

Mammalian expression vectors are engineered DNA molecules that deliver and control expression of selected genes in mammalian cells, enabling production or study of proteins in a biologically relevant cellular environment. They typically combine a mammalian promoter and regulatory elements with a coding sequence, termination or polyadenylation signal, and often a selectable marker; after transfection, the vector supports transcription in the nucleus and translation of the resulting messenger RNA. Researchers use these vectors for transient or stable gene expression, recombinant protein production, functional studies, and development of cell-based assays. Their design influences expression level, duration, cellular specificity, and experimental reproducibility.

Expression Analysis of Mammalian Linker-histone Subtypes

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.
