Map Kinase Pathway

The MAP kinase pathway is a conserved cell-signaling network that converts external stimuli into changes in cellular activity, including gene expression, growth, and survival. Typically, an activated receptor or upstream signaling protein initiates a phosphorylation cascade in which a MAP kinase kinase kinase activates a MAP kinase kinase, which then activates a MAP kinase. The activated MAP kinase phosphorylates cytoplasmic proteins and transcription factors, linking signals such as growth factors or stress to coordinated cellular responses. Studying this pathway helps explain development, immune responses, and disease mechanisms, including how abnormal signaling can contribute to cancer and other disorders.

This protocol describes an optogenetic strategy to modulate mitogen-activated protein kinase (MAPK) activity during cell differentiation and Xenopus embryonic development.
