Membrane Labeling

Membrane labeling is a set of methods used to mark cell membranes for visualization, identification, and analysis in biological research. Labels may consist of fluorescent dyes, lipid-conjugated probes, or tagged membrane proteins that associate with specific membrane components, allowing researchers to track membrane location, organization, movement, and interactions under suitable imaging conditions. These approaches support studies of cell structure, signaling, trafficking, adhesion, and membrane dynamics, while enabling techniques such as fluorescence microscopy and live-cell imaging to examine how membranes change during development, cellular communication, infection, or disease.

Examining the Conformational Dynamics of Membrane Proteins in situ with Site-directed Fluorescence Labeling

We will describe a method which measures the kinetics of ion transport of membrane proteins alongside site-specific analysis of conformational changes using fluorescence on single cells. This technique is adaptable to ion channels, transporters and ion pumps and can be utilized to determine distance constraints between protein subunits.

Single-Molecule Localization Microscopy of Membrane Proteins using Single-Antibody Labeling

This protocol describes single-antibody labeling (SAL) to resolve the nanoscale spatial organization of plasma membrane proteins. By leveraging cumulative antibody-epitope interactions at the single-molecule level, membrane SAL (mSAL) maps local epitope distributions while simultaneously capturing antibody binding behavior in the native cellular environment.

Metabolic Labeling and Membrane Fractionation for Comparative Proteomic Analysis of Arabidopsis thaliana Suspension Cell Cultures

Here we describe a robust method for the fractionation of plant plasma membranes into detergent resistant and detergent soluble membranes based on a mixture of unlabeled and in vivo fully 15N labeled Arabidopsis thaliana cell cultures. The procedure is applied for comparative proteomic studies to understand signaling processes.

Use of a Robot for High-throughput Crystallization of Membrane Proteins in Lipidic Mesophases

Herein is described a robotic approach to high-throughput crystallization of membrane proteins in lipidic mesophases for use in structure determination using macromolecular X-ray crystallography. Three robots capable of handling the viscous and sticky protein-laden mesophase integral to the method are introduced.
