Microscopic Analysis

Microscopic analysis is the systematic examination of cells, tissues, microorganisms, and other biological specimens with microscopes to reveal structures too small to see unaided. It works by using lenses or electron beams to magnify specimens, while illumination, staining, and contrast methods distinguish components and improve resolution. In biology, microscopic analysis supports cell identification, tissue characterization, microbial observation, and investigation of processes such as cell division and disease-related structural changes. By linking visible form with biological function, it provides essential evidence for research, diagnosis, teaching, and the development of new imaging methods.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.

Extracellular vesicles (EVs) contribute to cellular biology and intercellular communications. There is a need for practical assays to visualize and quantify EVs uptake by the cells. The current protocol proposes the EV uptake assay by utilizing three-dimensional fluorescence imaging via confocal microscopy, following EV isolation by a nano-filtration-based microfluidic device.
