Microwell Array Coating

Microwell array coating is a surface-engineering technique that modifies the interior of regularly patterned microwells to control how biological cells interact with and organize within them. Coatings such as extracellular matrix proteins can promote cell adhesion, while nonadhesive materials can limit attachment to defined regions, enabling reproducible cell loading, positioning, and aggregation. In biology, coated microwell arrays support the formation of uniform spheroids and organoid-like cultures, where controlled cell-cell interactions improve consistency across experiments. This approach is useful for studying development, tissue organization, disease models, and drug responses while reducing variability in three-dimensional cell culture.

2025

Source: Fattahi, N., et al. Photodegradable Hydrogel Interfaces for Bacteria Screening, Selection, and Isolation. J. Vis. Exp. (2021)This video demonstrates the use of photodegradable hydrogels to isolate bacterial colonies within silicon microwell arrays. Bacterial cells are first loaded into microwells, encapsulated in a crosslinked hydrogel, and cultured into distinct colonies. Upon exposure to ultraviolet light, targeted degradation of the hydrogel enables selective extraction of individual...

The development of microbial communities depends on a combination of factors, including environmental architecture, member abundance, traits, and interactions. This protocol describes a synthetic, microfabricated environment for the simultaneous tracking of thousands of communities contained in femtoliter wells, where key factors such as niche size and confinement can be approximated.

This manuscript introduces a robust method of fabricating concave microwells without the need for complex high-cost facilities. Using magnetic force, steel beads, and a through-hole array, several hundred microwells were formed in a 3 cm x 3 cm polydimethylsiloxane (PDMS) substrate.

In Vivo Real-Time Glutamate Monitoring Using an Enzyme-Coated Microelectrode Array

In this video, a Multielectrode Array (MEA) coated with glutamate Oxidase is implanted into the brain tissue of an anesthetized mouse. Glutamate solution is injected into the extracellular space where the MEA detects the glutamate levels, enabling real-time monitoring of glutamate uptake by the astrocytes.
