Intestinal Uptake

Intestinal uptake is the biological process by which nutrients, water, and other substances move from the gut lumen across the intestinal epithelium into the circulation or lymphatic system. It occurs through mechanisms such as passive diffusion, transporter-mediated movement, and vesicular uptake, with absorption shaped by molecular properties, epithelial barriers, and local conditions. In biology, studying intestinal uptake helps explain nutrient availability, metabolic regulation, and interactions between the gut and the rest of the body. The same principles guide research on oral drug delivery, bioavailability, dietary compounds, and intestinal disease, where altered epithelial transport can affect health and treatment outcomes.

M cells in a specialized follicle-associated epithelium covering Peyer’s patches play an important role for the mucosal immunosurveillance in gut-associated lymphoid tissue. Here we described the evaluation method for bacterial transcytosis by M cells in vivo. This method provides a method to understand M-cell function in the immune system.

In this video protocol we give a step by step explanation of lentiviral transduction in organoids of primary intestinal epithelium and of processing and downstream analysis of these cultures by quantitative RT-PCR, RNA-microarray and immunohistochemistry.

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

2025

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

Oral Intubation of Adult Zebrafish: A Model for Evaluating Intestinal Uptake of Bioactive Compounds

The protocol describes intubating adult zebrafish with a biologic; then dissecting and preparing the intestine for cytometry, confocal microscopy and qPCR. This method allows administration of bioactive compounds to monitor intestinal uptake and the local immune stimulus evoked. It is relevant for testing the intestinal dynamics of oral prophylactics.

Mitochondrial Calcium Uptake Assay: A Plate Reader-Based Method for Measuring Calcium Uptake in Isolated Mitochondria
