Neomycin Resistance Selection

Neomycin resistance selection is a laboratory method for identifying cells that carry a genetic construct containing a neomycin-resistance marker. The marker usually encodes neomycin phosphotransferase, an enzyme that phosphorylates aminoglycoside antibiotics and prevents them from disrupting bacterial or eukaryotic ribosomes; cells lacking the marker are inhibited or die under antibiotic exposure. Researchers apply neomycin or related compounds such as kanamycin or G418 to culture populations after transformation or transfection, enriching for resistant cells. This approach supports plasmid maintenance, stable cell-line generation, gene-function studies, and selection of successfully engineered organisms.
