Neural Stem Cell Lineage

Neural stem cell lineage describes the progression of neural stem cells through self-renewal, progenitor states, and differentiation into neurons and glial cells, making it central to nervous-system development and tissue maintenance. In response to signals from the cellular niche, neural stem cells divide symmetrically or asymmetrically, while transcriptional programs and environmental cues guide progenitors toward neuronal, astrocytic, or oligodendrocytic fates. Studying these transitions with lineage tracing, cell culture, and molecular profiling helps researchers define how neural circuits form and how stem-cell behavior changes after injury or in disease. This knowledge supports research into brain development, neurodegeneration, and regenerative strategies.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. This equipment meets clinical good manufacturing practice (cGMP) and clinical good lab practice (cGLP) guidelines.

Neural crest (NC) cells derived from human pluripotent stem cells (hPSC) have great potential for modeling human development and disease and for cell replacement therapies. Here, a feeder-free adaptation of the currently widely used in vitro differentiation protocol for the derivation of NC cells from hPSCs is presented.
