Neurocircuit Assay Protocol

A Neurocircuit Assay Protocol is a standardized method for measuring how interconnected neurons communicate and generate coordinated activity, helping link cellular function to behavior and disease. The approach typically applies a defined stimulus or perturbation to selected neural components, then records circuit responses through measures such as electrophysiology, fluorescence-based activity imaging, or behavioral output. By comparing responses across experimental conditions, researchers can assess synaptic connectivity, signal processing, and circuit-level effects of genetic, pharmacological, or environmental changes. These assays support studies of nervous system development, neurological disorders, drug action, and the relationship between neural activity and behavior.

Reverse genetic approaches have proven extremely useful for determining which genes underly resistance to vector pathogens in mosquitoes. This video protocol illustrates a method used by the Dimopoulos lab to inject dsRNA into Anopheles gambiae mosquitoes, which harbor the malaria parasite. The technique manipulating the injection setup and injecting dsRNA into the thorax is illustrated.

Protein Membrane Overlay Assay: A Protocol to Test Interaction Between Soluble and Insoluble Proteins in vitro

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
