Nipple Injection Technique

Nipple injection technique is a localized method for introducing experimental substances into the mammary gland through the nipple, enabling direct access to the ductal system in biological research. A fine needle or cannula enters the nipple opening, and controlled delivery distributes a solution through the mammary ducts while limiting exposure to surrounding tissues. Researchers use this approach to investigate mammary gland development, epithelial biology, tumor formation, and responses to candidate treatments. Because it targets a defined anatomical compartment, the technique can improve experimental precision and support studies of mammary tissue organization and function.

Intraductal Injection for Localized Drug Delivery to the Mouse Mammary Gland

A protocol for the non-invasive intraductal delivery of aqueous reagents to the mouse mammary gland is described. The method takes advantage of localized injection into the nipples of mammary glands targeting mammary ducts specifically. This technique is adaptable for a variety of compounds including siRNA, chemotherapeutic agents and small molecules.

Repair of the intracarotid artery in a mouse model after injection returns blood flow to the artery without negatively impacting the distribution of the injected material. Injection site repair facilitates subsequent injections through the same artery and prevents cerebral ischemia in mouse strains that lack a complete Circle of Willis.
