Non-cleavable Internal Import Signals

Non-cleavable internal import signals are amino acid sequences embedded within a protein that direct its transport to a specific cellular compartment without being removed after import. Unlike cleavable targeting peptides at a protein terminus, these signals remain part of the mature protein and are recognized by organelle-specific receptors and membrane translocation machinery, often through their sequence, charge, and hydrophobic properties. In cell biology, they help regulate the localization and function of proteins in organelles such as mitochondria, peroxisomes, and the endoplasmic reticulum. Studying these signals clarifies protein trafficking and supports research on organelle biogenesis, cellular organization, and disease-associated targeting defects.
