Tom20-tom22 Receptor Complexes

Tom20-Tom22 receptor complexes are protein assemblies in the mitochondrial outer membrane that recognize incoming precursor proteins and initiate their import, a process essential for mitochondrial biogenesis and function. Tom20 binds mitochondrial targeting signals, while Tom22 contributes to receptor organization and helps guide selected precursors toward the TOM40 translocation channel, where they cross the outer membrane. Studying this receptor complex clarifies how proteins are sorted to mitochondria, how defects can disrupt cellular energy metabolism, and how mitochondrial protein trafficking is regulated in health and disease. Its structure and activity also provide a foundation for investigating organelle assembly and mitochondrial disorders.

An in vitro method for preparing functional glucocorticoid receptor (GR)•hsp90 protein complexes from purified proteins and cellular lysates is described. The method utilizes immunoadsorption of recombinant GR followed by salt-stripping and protein complex reconstitution. The importance of cofactors and buffer conditions are discussed, as are potential method applications.

Olfactory receptor activation patterns encode odor identity, but the lack of published data identifying odorant ligands for mammalian olfactory receptors hinders the development of a comprehensive model of odor coding. This protocol describes a method to facilitate high-throughput identification of olfactory receptor ligands and quantification of receptor activation.
