Nuclear Import

Nuclear import is the selective transport of proteins, RNA-protein complexes, and other macromolecules from the cytoplasm into the cell nucleus, where it supports gene regulation, DNA replication, and genome maintenance. Cargo proteins containing a nuclear localization signal are recognized by importin receptors, which carry them through nuclear pore complexes; inside the nucleus, Ran-GTP binds importin and releases the cargo, while the Ran-GTPase cycle drives receptor recycling and directionality. Studying nuclear import helps explain how cells control transcription and signaling, and it provides insight into viral infection, developmental disorders, cancer, and strategies for delivering therapeutic molecules to the nucleus.

Proteins targeted to the nucleus carry short stretches of amino acid sequences called the nuclear localization signal or NLS. We provide a detailed protocol for induction of long-term potentiation in the CA1 region of the hippocampus and the subsequent isolation of nuclear enriched fractions from the tetanized area of the slice. This approach can be used to determine activity dependent nuclear protein import in cellular models of learning and memory.

Here we describe a new method to study protein import into isolated chloroplasts under stress. The method is rapid and straightforward, and can be applied to study the consequences of different stress conditions for chloroplast protein import, and the corresponding regulatory mechanisms.
