Drosophila Larvae Dissection

Drosophila larvae dissection is a laboratory method for opening and examining the internal tissues and organs of fruit fly larvae, making developmental biology visible at cellular and tissue levels. In practice, researchers immobilize a larva, make a controlled opening in its cuticle, and use saline and microscopy to expose, maintain, and inspect structures such as the nervous system, muscles, gut, and imaginal discs. This preparation supports studies of organ development, cell differentiation, gene function, and physiological responses while enabling direct comparison of normal and experimentally altered tissues.

This protocol demonstrates the dissection technique used for isolating imaginal discs from drosophila larvae at the 3rd instar stage. Methods for fixing the tissue after saying and removing the wing, leg, and eye discs are demonstrated directly on a microscope slide for subsequent visualization.

Drosophila Larva Imaginal Disc Dissection: A Method to Observe Developing Epithelia

When staining Drosophila imaginal discs, often a rough dissection is performed where the discs are exposed but remain attached to the body. Once stained, the discs of interest are isolated and mounted. The example protocol demonstrates a procedure for wing imaginal disc dissection and mounting.

Drosophila melanogaster Embryo and Larva Harvesting and Preparation

2023

Drosophila melanogaster embryos and larvae are easy to manipulate and develop rapidly by mechanisms that are analogous to other organisms, including mammals. For these reasons, many researchers utilize fly embryos and larvae to answer questions in diverse fields ranging from behavioral to developmental biology. Prior to experimentation, however, the embryos and larvae must first be collected. This video will first demonstrate how "egg-laying cups" are used to collect Drosophila embryos on agar...
