Nuclear Strain Analysis

Nuclear strain analysis is a quantitative method for measuring how cell nuclei deform when exposed to mechanical forces, making nuclear mechanics accessible in biological research. By comparing nuclear shape, position, or displacement under defined conditions, researchers can estimate strain and relate it to force transmission from the cytoskeleton through the LINC complex and nuclear envelope to chromatin. This approach helps investigate mechanotransduction, cell migration, tissue organization, and changes in gene regulation associated with development or disease. In biology, nuclear strain analysis links measurable physical deformation with cellular responses, supporting studies of mechanobiology and the effects of altered mechanical environments.

We illustrate the application of 1H(15N,αγ)12C resonant nuclear reaction analysis (NRA) to quantitatively evaluate the density of hydrogen atoms on the surface, in the volume, and at an interfacial layer of solid materials. The near-surface hydrogen depth profiling of a Pd(110) single crystal and of SiO2/Si(100) stacks is described.
