Human Vascular Network

The human vascular network is the interconnected system of blood vessels that circulates blood throughout the body, delivering oxygen and nutrients while removing metabolic waste. The heart drives blood through arteries and arterioles to capillaries, where thin vessel walls permit exchange with tissues; blood then returns through venules and veins. This network also helps regulate blood pressure, temperature, and tissue homeostasis through changes in vessel diameter and blood flow. Studying vascular structure and function supports research into cardiovascular disease, tumor growth, wound healing, and the development of treatments that restore or control blood supply.

This protocol outlines the implementation of image-guided, laser-based hydrogel degradation to fabricate vascular-derived, biomimetic microfluidic networks embedded in poly(ethylene glycol) diacrylate (PEGDA) hydrogels. These biomimetic microfluidic systems may be useful for tissue engineering applications, generation of in vitro disease models, and fabrication of advanced "on-a-chip" devices.

Bioengineering Human Microvascular Networks in Immunodeficient Mice

Here, we describe a methodology to deliver human cord blood-derived endothelial colony-forming cells (ECFCs) and bone marrow-derived mesenchymal stem cells (MSCs), embedded in a collagen/fibronectin gel, subcutaneously into immunodeficient mice. This cell/gel combination generates a human vascular network that connects with the mouse vasculature.

The protocol describes how to engineer a perfusable vascular network in a spheroid. The spheroid's surrounding microenvironment is devised to induce angiogenesis and connect the spheroid to the microchannels in a microfluidic device. The method allows the perfusion of the spheroid, which is a long-awaited technique in three-dimensional cultures.
