Hepatic Endoderm

Hepatic endoderm is the population of embryonic endodermal cells that gives rise to the liver and parts of the biliary system, making it essential for understanding organ formation and liver disease. During development, foregut endoderm receives inductive signals, including fibroblast growth factor, bone morphogenetic protein, and Wnt pathways, from neighboring tissues; these cues specify hepatic identity, trigger liver bud formation, and generate proliferative hepatoblasts. Hepatoblasts then differentiate into hepatocytes or cholangiocytes as local signals and changing tissue environments guide maturation and organization. Research on hepatic endoderm supports studies of liver development, congenital disorders, organoid formation, drug testing, and regenerative medicine.

Generation of Functional Endodermal Hepatic Organoids

This protocol describes the generation of fast and reproducible endodermal hepatic organoids (eHEPOs). With this protocol, eHEPOs can be produced within 2 weeks and expand long-term (more than 1 year) without losing their differentiation and functionality.

An optical system was developed to visualize hepatic microcirculation with FITC-labeled erythrocytes and to measure the partial pressure of oxygen in the microvessels with laser-assisted phosphorimetry. This method can be used to investigate physiological and pathological mechanisms by analyzing microvascular structure, diameter, blood flow velocity, and oxygen tension.
