Nucleotide Pool Dynamics

Nucleotide pool dynamics describes how cells regulate the abundance, composition, and turnover of nucleotide metabolites, which supply RNA and DNA synthesis and support cellular energy transfer and signaling. Through coordinated de novo synthesis, salvage, phosphorylation, degradation, and compartmentalized transport, enzymes maintain balanced ribonucleotide and deoxyribonucleotide pools; feedback control adjusts pathway activity in response to growth, nutrient availability, and DNA replication demand. Disrupted pool size or composition can stall replication, impair repair, and increase mutagenesis, making nucleotide pool dynamics important in genetics, cancer biology, antimicrobial research, and drug development. Measuring these changes helps connect metabolic regulation with genome stability and can reveal therapeutic vulnerabilities.

Detection of Rare Genomic Variants from Pooled Sequencing Using SPLINTER

Pooled DNA sequencing is a fast and cost-effective strategy to detect rare variants associated with complex phenotypes in large cohorts. Here we describe the computational analysis of pooled, next-generation sequencing of 32 cancer-related genes using the SPLINTER software package. This method is scalable, and applicable to any phenotype of interest.

Pool-boiling heat-transfer experiments were carried out to observe the effects of hybrid wettable patterns on the heat-transfer coefficient (HTC). The parameters of investigation are the number of interlines and the pattern orientation of the modified wettable surface.
