Oct Embedding

OCT embedding is a biological sample-preparation method that encases tissue in optimal cutting temperature compound for frozen-section analysis. The specimen is oriented in the water-soluble medium, rapidly frozen, and cut into thin sections with a cryostat; the solidified compound supports the tissue while preserving its structure and temperature-sensitive components. These sections can be placed on microscope slides for histological staining, immunohistochemistry, fluorescence imaging, or molecular analysis. OCT embedding is especially useful when researchers need to examine tissue architecture, localize proteins or cells, or analyze samples that may be altered by chemical fixation and paraffin processing.

Plastic sections maintain true tissue morphology in thin sections of tissue that can be immunostained with fluorescent secondary antibodies, making this method more useful than paraffin-embedded or frozen sections for many types of tissue. The method for staining, plastic embedding, and sectioning is demonstrated in this video.
