Opc Differentiation

OPC differentiation is the biological process by which oligodendrocyte precursor cells transition from proliferative, migratory progenitors into mature oligodendrocytes, the glial cells that produce myelin in the central nervous system. During this transition, OPCs withdraw from the cell cycle, alter gene expression, extend complex processes, and establish contact with axons before forming myelin sheaths. The process is regulated by developmental signals, neuronal activity, and the local tissue environment. Studying OPC differentiation helps explain nervous system development, axon insulation, and repair after demyelinating injury, while supporting research into multiple sclerosis, spinal cord damage, and remyelination-based therapies.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.

The video demonstrates the differentiation of immortalized multipotent otic progenitors (iMOPs) into spiral ganglion neurons (SGNs) and immunofluorescence-based confirmation of the differentiation. The iMOPs are plated onto culture substrate-coated coverslips in a neuronal differentiation medium for differentiation into SGNs. The differentiated cells are labeled with antibodies specific for neuronal differentiation markers and analyzed under a microscope to confirm differentiation.
