Pbs Flush

PBS flush is a laboratory rinsing step that uses phosphate-buffered saline (PBS) to remove residual blood, culture medium, cells, or other soluble contaminants while helping preserve biological samples. Its action depends on controlled flow through or across a sample, with phosphate buffering limiting pH changes and sodium chloride and related salts maintaining near-isotonic conditions that reduce osmotic stress. In biology, researchers use PBS flushes during tissue perfusion, sample preparation, cell handling, and equipment or vessel rinsing. Consistent flushing improves cleanliness and reproducibility, supporting microscopy, immunostaining, molecular assays, and downstream analysis.

This video demonstrates the size exclusion chromatography (SEC) technique for purifying bacterial extracellular vesicles (EVs) from the culture filtrate of Mycobacterium tuberculosis (Mtb). The technique utilizes a porous resin-packed column to obtain purified fractions of EVs. The porous particles selectively allow smaller proteins to enter the pores, resulting in slower elution. At the same time, EVs are excluded from entering the pores due to their larger size and are eluted first.
