Peptide Loaded Mhc

Peptide-loaded major histocompatibility complex (MHC) molecules are cell-surface protein complexes that display short peptide fragments for recognition by T cells. They form when antigen-processing pathways generate peptides that bind within an MHC groove, allowing the complex to be transported to the cell surface, where its peptide is examined by a T-cell receptor. MHC class I primarily presents intracellular peptides to CD8+ T cells, whereas MHC class II presents peptides from extracellular materials to CD4+ T cells. Studying peptide-loaded MHC supports understanding of immune surveillance, infection, autoimmunity, and cancer, and guides vaccine design, antigen-specific assays, and immunotherapies.

A High Throughput MHC II Binding Assay for Quantitative Analysis of Peptide Epitopes

Biochemical assays with recombinant human MHC II molecules can provide rapid, quantitative insights into immunogenic epitope identification, deletion, or design. Here, a peptide-MHC II binding assay scaled to 384-well plates is described. This cost effective format should prove useful in the fields of protein deimmunization and vaccine design and development.

This video demonstrates a method for characterizing CD8+ memory T cells specific to simian immunodeficiency virus (SIV) using peptide-major histocompatibility complex class I (pMHC-I) tetramers. A pMHC-I tetramer is a complex of four MHC-I molecules bound to SIV-specific peptides and conjugated to a fluorophore. These tetramers selectively bind to T cell receptors (TCRs) on SIV-specific CD8+ T cells, facilitating the characterization of these cells.
