Alan Doucette

Dalhousie University, Department of Chemistry

Dr. Alan Doucette is a Professor of Chemistry at Dalhousie University, in Halifax, Canada, a place he is proud to call home, as it was here that he earned his BSc degree. His PhD work at the University of Alberta (’97 under supervisor Dr. Liang Li) motivated his current research, which centers on the development of new technologies to improve proteome characterization by mass spectrometry (MS). Doucette focuses on the creation of front-end sample preparation devices that enable the manipulation of proteins, particularly for top-down proteoform analysis. Highlights of his past work include the GELFrEE protein separation platform, which exploits the benefits of SDS to enable mass-based proteome fractionation. SDS depletion and purification can be achieved through protein precipitation, as Doucette has shown that the combination of salt and organic solvents achieves exceptional high recovery. Precipitation is simplified with the commercial ProTrap XG, another commercial example from the Doucette lab. Alan is also passionate about chemical education and inspiring the next generation of researchers.

Collection Overview

With improved speed, sensitivity, and resolution, advances in mass spectrometry instrumentation have revolutionized the field of proteomics. With this improved technology, researchers finally have a tool sufficient to address important biological questions – or do they? Mass spectrometry is but one aspect of the complex proteome workflow, which begins from the moment of sample collection, and carries through to the proper interpretation of MS data (which tends to spark the next study). This focused issue sheds light on the many aspects surrounding MS analysis of complex biological systems. Possible topics include strategies that improve the consistency of front-end protein sample preparation (e.g. protein isolation, modification and separation), interfacing to MS, accelerated approaches for MS data acquisition, and methods for data interpretation, storage, and statistical analysis, including experimental design.
