Perfusion Culture

Perfusion culture is a continuous cell-culture method in which fresh growth medium flows through a bioreactor while spent medium and metabolic waste are removed, enabling cells to remain in a controlled environment for extended periods. A perfusion system maintains nutrient supply and environmental conditions by regulating flow, often using a retention device that keeps cells inside the vessel while allowing dissolved nutrients and waste products to pass through. This approach supports high cell densities and sustained production of biologics, including antibodies and recombinant proteins. In biology, perfusion culture also helps model tissue environments and study cell growth, differentiation, and responses under more stable conditions than batch culture.

Chip-based Three-dimensional Cell Culture in Perfused Micro-bioreactors

We describe a chip-based platform for the three-dimensional cultivation of cells in micro-bioreactors. One chip can house up to 10 Mio. cells that can be cultivated under precisely defined conditions with regard to fluid flow, oxygen tension etc. in a sterile, closed circulation loop.

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

Langendorff Retrograde Heart Perfusion: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.
