Periosteum

The periosteum is a specialized, vascular connective-tissue membrane that covers the external surfaces of most bones, providing structural support, sensation, and a living interface for bone growth and repair. It contains an outer fibrous layer and an inner cellular, osteogenic layer; collagenous Sharpey’s fibers anchor it to bone, while blood vessels, nerves, and progenitor cells help coordinate nutrient delivery, pain signaling, and new bone formation. In biology and medicine, the periosteum is central to understanding skeletal development, fracture healing, bone graft integration, and orthopedic procedures, where preserving or stimulating its activity can improve tissue regeneration and functional recovery.

We present a protocol to investigate the mRNA expression biomarkers of periosteum-derived cells (PDCs) induced by vitamin C (vitamin C) and 1,25-dihydroxy vitamin D [1,25-(OH)2D3]. In addition, we evaluate the ability of PDCs to differentiate into osteocytes, chondrocytes, and adipocytes.
