Phosphatidylinositol Triphosphate

Phosphatidylinositol triphosphate, commonly called phosphatidylinositol (3,4,5)-trisphosphate or PIP3, is a signaling lipid that helps cells translate extracellular cues into changes in growth, survival, metabolism, and movement. At the plasma membrane, phosphoinositide 3-kinase (PI3K) produces PIP3 by adding a phosphate group to phosphatidylinositol (4,5)-bisphosphate, while phosphatases such as PTEN remove it to limit signal duration. PIP3 recruits proteins containing pleckstrin homology domains, including AKT and PDK1, positioning them for activation and downstream signaling. Studying this pathway supports research on insulin responses, cell proliferation, development, and diseases such as cancer.

PI(4,5)P2 regulates various cellular functions, but its nanoscale organization in the cell plasma membrane is poorly understood. By labeling PI(4,5)P2 with a dual-color fluorescent probe fused to the Pleckstrin Homology domain, we describe a novel approach to study the PI(4,5)P2 spatial distribution in the plasma membrane at the nanometer scale.

We present a cost-effective method for assessing liposomal membrane permeability with superior sensitivity and resolution over conventional techniques. Using a novel encapsulated ligand substitution reaction, this technique detects phospholipase A2-induced changes in model membranes. Easily integrated with standard analyses, this accessible approach enhances resolution for studying membrane biophysics and toxin interactions.

This video demonstrates a technique for detecting local and global calcium ion response upon Shigella infection of human epithelial cells. Shigella invades host cells and releases invasion effectors, impacting intracellular calcium levels. The amplitude and frequency of the local and global cytosolic calcium are studied using a fluorescent indicator.
