Phospholipase Treatment

Phospholipase treatment uses phospholipase enzymes to selectively hydrolyze phospholipids, the major structural lipids in biological membranes, and is important for studying membrane composition and lipid signaling. Depending on the enzyme, phospholipase activity cleaves specific bonds within a phospholipid, producing products such as lysophospholipids, fatty acids, or phosphorylated head groups; reaction conditions and substrate accessibility influence the outcome. In biology, this treatment helps identify membrane lipids, investigate membrane remodeling and permeability, release signaling molecules, and prepare samples for biochemical analysis. It can also clarify how lipid composition affects cellular structure and function.

In this video, we demonstrate an assay to establish the presence of phospholipase in the crude venom extract from Anthopleura dowii—a marine sea anemone. The assay utilizes an agarose gel diffusion technique, in which the enzyme phospholipase diffuses and breaks down phospholipids incorporated in the solid media in a radial manner.

To test the inhibitory effects of pharmacologic agents on phospholipase C (PLC) in different regions of the honeybee brain, we present a biochemical assay to measure PLC activity in those regions. This assay could be useful for comparing PLC activity among tissues, as well as among bees exhibiting different behaviors.
