Photon Flux Quantification

Photon flux quantification is the measurement of the number of photons reaching a surface per unit area and time, a key parameter for describing light exposure in biological systems. Instruments such as quantum sensors or calibrated spectroradiometers detect incident radiation and convert it into photon-based measurements, often accounting for wavelength because photon energy varies across the spectrum. In biology, these measurements standardize illumination for photosynthesis experiments, plant growth studies, circadian research, microscopy, and photobiology. Accurate photon flux data improves reproducibility, enables comparisons between light treatments, and helps researchers relate cellular or organismal responses to defined optical conditions.

By employing a spectrally resolved two-photon microscopy imaging system, pixel-level maps of Förster Resonance Energy Transfer (FRET) efficiencies are obtained for cells expressing membrane receptors hypothesized to form homo-oligomeric complexes. From the FRET efficiency maps, we are able to estimate stoichiometric information about the oligomer complex under study.

The presented protocol uses the eddy covariance method at non-typical locations, applicable to all types of short-canopy ecosystems with limited area, on a currently reforested windthrow site in Poland. Details of measuring site setup rules, flux calculations and quality control, and final result analysis, are described.
