Plant Biomass Characterization

Plant biomass characterization is the systematic analysis of plant-derived material to determine its physical, chemical, and structural properties, supporting research in biology, agriculture, and bioenergy. It commonly combines measurements of moisture and ash with compositional analysis of structural carbohydrates, cellulose, hemicellulose, lignin, proteins, and other constituents. Researchers use techniques such as gravimetric assays, spectroscopy, microscopy, and elemental analysis to relate biomass composition and structure to its origin, quality, digestibility, and processing behavior. These measurements help assess plant growth and tissue composition, compare feedstocks, guide conversion into fuels and bioproducts, and evaluate biomass for ecological and industrial applications.

This video demonstrates an assay to screen enzymes effective in degrading complex polysaccharides in plant biomass using insoluble chromogenic biomass or ICB. The enzymes hydrolyze ICB to release soluble dye-linked oligosaccharides, and the enzyme activity is assessed by colorimetric analysis of the reaction product.

Estimation of Plant Biomass Lignin Content using Thioglycolic Acid (TGA)

Here, we present a modified TGA method for estimation of lignin content in herbaceous plant biomass. This method estimates the lignin content by forming specific thioether bonds with lignin and presents an advantage over the Klason method, as it requires a relatively small sample for lignin content estimation.
