Pre-embedding Protocol

A pre-embedding protocol comprises the preparation steps performed on a biological specimen before it is enclosed in a supporting medium for microscopy, preserving structure while enabling targeted analysis. Depending on the specimen and imaging method, the workflow may include fixation, permeabilization, labeling with antibodies or other probes, dehydration, and infiltration before embedding in resin or another matrix. These steps help preserve cellular architecture and position molecular markers for visualization, particularly in electron microscopy and immunolabeling studies. Careful control of reagent exposure and processing conditions improves label penetration, structural preservation, and the reliability of downstream ultrastructural or localization analyses.
