Epithelial Stem Cells

Epithelial stem cells are self-renewing cells that maintain and repair epithelial tissues, the protective layers lining surfaces such as skin, intestine, and airways. Through asymmetric or symmetric division, they replenish stem and progenitor cell populations, while signals from the surrounding niche regulate proliferation, differentiation, and tissue organization. In biology, studying these cells clarifies how tissues develop, preserve barrier function, and recover after injury, while also revealing mechanisms involved in cancer and chronic disease. Their controlled renewal supports organoid research, disease modeling, regenerative medicine, and therapies designed to restore damaged epithelia.

Rapid, Directed Differentiation of Retinal Pigment Epithelial Cells from Human Embryonic or Induced Pluripotent Stem Cells

This protocol describes how to produce retinal pigment epithelial cells (RPE) from pluripotent stem cells. The method uses a combination of growth factors and small molecules to direct the differentiation of stem cells into immature RPE in fourteen days and mature, functional RPE after three months.

A rapid, robust way of isolating viable adult epithelial stem cells from human skin is described. The method utilizes enzymatic digestion of skin collagen matrix , followed by plucking of hair follicles and isolation of single cell suspensions or tissue fragments for cell culture.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
