Pre-mirna

Pre-miRNA is a short, hairpin-shaped RNA intermediate in microRNA biogenesis, the pathway that produces small regulatory RNAs controlling gene expression. It forms when the Microprocessor complex, chiefly Drosha and DGCR8, cleaves a longer primary microRNA transcript in the nucleus; Exportin-5 then transports the precursor to the cytoplasm, where Dicer cuts it into a short RNA duplex. One strand is loaded into the RNA-induced silencing complex and guides sequence-specific recognition of target messenger RNAs, reducing their translation or stability. Studying pre-miRNA processing clarifies post-transcriptional regulation and supports research on development, disease mechanisms, biomarkers, and RNA-based therapies.

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.

In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA.
