Preclinical Drug Assessment

Preclinical drug assessment is the systematic evaluation of a candidate therapy before testing in humans, used to determine whether it has sufficient biological activity and an acceptable safety profile. Researchers combine cell-based and biochemical assays with animal studies to measure pharmacodynamics, pharmacokinetics, dose-response relationships, toxicity, and how the body absorbs, distributes, metabolizes, and eliminates the compound. In biology, these findings guide lead optimization and selection of development candidates, identify potential risks and mechanisms of action, and provide evidence for designing carefully monitored clinical trials.

Implementation of an orthotopic model of renal cell carcinoma in immunocompetent mice affords the investigator a clinically-relevant system defined by the presence of a primary renal tumor and lung metastases in the same animal. This system can be used to preclinically test a variety of treatments in vivo.

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.
