Pupal Stage Development

Pupal stage development is the transitional phase in the life cycle of holometabolous insects, during which a larva is reorganized into an adult and major body structures form. After hormonal signals, including a rise in ecdysone and changes in juvenile hormone, initiate pupation, larval tissues break down while imaginal discs and other progenitor cells develop adult features such as wings, legs, and reproductive organs. Studying this stage helps explain insect metamorphosis, growth regulation, and developmental timing. It also supports research in developmental biology, species identification, ecology, and pest-management strategies that target insects before adulthood.

Insect Puparium Removal: A Method for Pupal Investigation

This article describes a technique to remove an insect’s puparium while keeping the inside pupa intact and therefore fit for further investigation. The example protocol shows the procedure with Drosophila guttifera; however, this technique is applicable to other Drosophila species.
