Quorum Quenching

Quorum quenching is the disruption of bacterial cell-to-cell communication by interfering with quorum sensing, a process that coordinates group behaviors according to cell density. It works by degrading or modifying signaling molecules, blocking their synthesis or transport, or preventing them from binding to regulatory receptors, thereby reducing the expression of quorum-sensing-controlled genes. In biology, quorum quenching helps explain how microbial communities regulate biofilm formation, virulence, motility, and cooperative metabolism. Its mechanisms support research into anti-biofilm strategies and alternatives to conventional antibiotics, with potential applications in medicine, environmental biotechnology, food safety, and agriculture.

Quorum-quenching enzymes are anti-virulent and anti-bacterial options that can mitigate pathogenesis without risk of incurring resistance, by preventing the expression of virulence factors and genes associated with antibiotic resistance and biofilm formation. In this study, we report a method that demonstrates the efficacy of quorum-quenching enzymes in bacterial biofilm disruption.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
