Renal Collecting Ducts

Renal collecting ducts are tubular structures in the kidney that receive fluid from nephrons and adjust its final composition before urine leaves the kidney. Their epithelial cells regulate water and electrolyte movement in response to hormonal signals, especially antidiuretic hormone, which promotes aquaporin insertion into the cell membrane and increases water reabsorption along the medullary osmotic gradient. Collecting ducts also contribute to acid-base balance by secreting hydrogen ions and managing bicarbonate. Their coordinated activity concentrates urine, maintains blood osmolarity and volume, and provides a key model for studying kidney function, hypertension, dehydration, and disorders of water regulation.

Renal Tubule and Collecting Duct

The renal tubule is divided into three parts: the proximal convoluted tubule (PCT), the Loop of Henle (LOH), and the distal convoluted tubule (DCT). Proximal Convoluted Tubule (PCT): The PCT is the initial segment of the renal tubule, extending from the Bowman's capsule that encloses the glomerulus. Its convoluted structure and microvilli-lined cells increase the surface area for reabsorption. The PCT reabsorbs glucose, amino acids, sodium, and water from the filtrate, ensuring essential...

Arginine-vasopressin (AVP) controls fine-tuning of body water homeostasis through facilitating water reabsorption by renal principal cells. Here, we present a protocol for the cultivation of primary rat inner medullary collecting duct cells suitable for the elucidation of molecular mechanisms underlying AVP-mediated water reabsorption.

Reabsorption and Secretion in the DCT and Collecting Duct

The early phase of the DCT manages the reabsorption of approximately 10-15% of filtered water, 5–10% of filtered sodium, and 5–10% of filtered chloride. This process is facilitated by Na+–Cl− symporters in apical membranes and sodium-potassium pumps, as well as Cl− leakage channels in basolateral membranes. The early DCT also stands out as a site where parathyroid hormone (PTH) stimulates calcium reabsorption, depending on the body's requirements. The distal part of the DCT, along with the...

2016

Early stage hemodynamic dysfunction is critical to the development of kidney disease. Yet, detection methodologies are limited. Recent advances in sonography provide a noninvasive, accurate option for early detection of kidney injury. This study outlines a step-by-step, sonographic methodology for detecting kidney dysfunction using a drug-induced nephrotoxicity rat model.
