Repeat Expansion Disorders

Repeat expansion disorders are inherited genetic diseases caused by abnormal increases in the number of tandemly repeated DNA sequences within or near a gene, disrupting normal gene function. During DNA replication or repair, repetitive sequences can mispair and form looped structures, allowing the repeat tract to expand across generations; depending on its location and sequence, the expansion may silence a gene or produce toxic RNA or protein. These disorders include Huntington disease, fragile X syndrome, and myotonic dystrophy, and often show genetic anticipation, in which symptoms begin earlier or become more severe in successive generations. Studying repeat instability supports molecular diagnosis, counseling, and development of targeted therapies.
