Bone Core Sampling

Bone core sampling is a technique that removes a cylindrical section of bone for biological, histological, or diagnostic analysis while preserving relationships among mineralized tissue, marrow, and cellular structures. A trephine or similar coring instrument cuts through the bone, and the specimen is stabilized, processed, and examined using methods such as fixation, decalcification, sectioning, and microscopy. In biology, bone cores support studies of tissue architecture, remodeling, mineralization, marrow composition, and disease-related changes. They also provide material for evaluating skeletal development, implant integration, and responses to therapeutic interventions, linking microscopic structure with bone function and health.

A New Workflow for Sampling and Digitizing Increment Cores

We present a protocol to use 3D-printed mounts to fix increment cores in the field without the necessity of unpacking and gluing them on wooden mounts. The new GSC holder allows placing the cores in a core microtome to cut their surface and directly transfer them to digital image capturing.

We employed a geological (coring) sampling protocol to procure cortical bone specimens of uniform size for SRµCT experiments from the anterior aspect of human femora. This method is minimally destructive, efficient, results in cylindrical specimens that minimize imaging artifacts from irregular sample shapes and improves microarchitectural visualization and analysis.
