Resazurin Fluorometric Assay

The Resazurin Fluorometric Assay is a cell-based method for estimating metabolic activity, viability, and proliferation in biological samples. It relies on the reduction of resazurin, a blue, nonfluorescent dye, to resorufin, a pink, fluorescent product by metabolically active cells during incubation. Fluorescence intensity therefore provides a quantitative readout that can be compared across treatments, although assay conditions must be optimized to maintain a measurable response and avoid signal saturation. In biology, the assay supports studies of cell growth, toxicity, drug response, and microbial or mammalian metabolism, offering a scalable alternative to direct cell counting.

Most bacterial infections produce a biofilm. By virtue of their environment, biofilm associated bacteria are often phenotypically drug resistant. Novel antibacterial molecules that kill bacteria in biofilms are thus a high priority. We establish an assay to quickly screen for antimicrobial compounds that are effective at eradicating biofilms.

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.
