Resolving Gel Preparation

Resolving gel preparation is the process of making the separating layer of a polyacrylamide gel used in gel electrophoresis to analyze proteins by size. Acrylamide and bisacrylamide polymerize into a porous network, while the chosen gel concentration determines pore size; during electrophoresis, an electric field drives charged proteins through the matrix, allowing smaller molecules to migrate faster than larger ones. The resolving gel is typically cast beneath a stacking gel, which concentrates protein samples into narrow bands before separation. Careful preparation supports clear, reproducible protein profiles for molecular biology, biochemical research, and applications such as assessing protein expression or purity.

Our protocol demonstrates how to pour multiple protein gels at a time by recycling Invitrogen Nupage Novex minigel cassettes, and inexpensive materials purchased at a home improvement store. This economical and streamlined method includes a way to store the gels at 4°C for a few weeks. By re-using the plastic gel cassettes from commercially available gels, labs that run frequent protein gels can save significant costs and help the environment.
