Safety Catch Method

The Safety Catch Method is a strategy for selectively releasing a biological molecule from a solid support after purification, improving control over affinity-based isolation. It uses a safety-catch linker with a latent reactive group that remains chemically inert during immobilization, binding, and washing; a separate activation step converts the linker into a labile form that can be cleaved to liberate the target molecule. In biology, this approach supports the purification of peptides, proteins, and other biomolecules while reducing nonspecific release and harsh elution conditions. Its controlled chemistry is useful in proteomics, molecular biology, and the preparation of research-grade biological reagents.

We present protocols for the application of our targeted genetically-encoded calcium indicator (GECI) CatchER+ for monitoring rapid calcium transients in the endoplasmic/sarcoplasmic reticulum (ER/SR) of HEK293 and skeletal muscle C2C12 cells using real-time fluorescence microscopy. A protocol for the in situ Kd measurement and calibration is also discussed.
